摘要
目的研究胃泌素受体拮抗剂和CD28/CpGODN活化PBMC在体外对结肠癌细胞株SW480杀伤作用机理。方法用含共刺激活化PBMC或(和)CI-988(胃泌素受体拮抗剂)的100ml/L胎牛血清的RPMI1640培养基培养细胞,测生长曲线;MTT法检测活化细胞的体外淋巴细胞毒作用,并用电镜观察效应细胞杀伤的结肠癌细胞超微结构及用流式细胞仪检测结肠癌细胞的凋亡指数。结果胃泌素拮受体抗剂本身对SW480细胞有一定的杀伤作用(P<0.05),CD28/CpGODN共刺激活化PBMC在体外对结肠癌细胞株SW480杀伤作用明显(P<0.01),胃泌素受体拮抗剂与CD28/CpGODN共刺激活化PBMC合用,对SW480CEA的杀伤作用显著大于胃泌素受体拮抗剂本身对SW480细胞杀伤作用(P<0.01),亦大于单独应用CD28/CpGODN共刺激活化PBMC对SW480细胞的杀伤作用(P<0.05),胃泌素受体拮抗剂增加了SW480细胞对共刺激活化PBMC的敏感性。电镜结果显示,效应细胞作用24小时肿瘤细胞就部分发生坏死,部分肿瘤细胞可见凋亡。流式细胞仪检测效应细胞SW480CEA细胞72小时明显凋亡。结论胃泌素受体拮抗剂与共刺激活化PBMC联合应用,提高了CD28/CpGODN共刺激活化PBMC对结肠癌细胞SW480的靶向性杀伤作用。活化的淋巴细胞杀伤结肠癌细胞是通过坏死及凋亡实现的。
Objective To investigate the killing effect of costimulate activation PBMC with CD28 MoAb and CpG ODN on the human colorectal carcinoma cell lines (SW480) when combined with gastrin receptor antagonist (CI-988). in vitro. Methods The growth curve of tumor cells was observed when cultured with CD28/GpG ODN activated PBMC and CI - 988. The killing efficiency was measured by MTY method. The submicroscopic structure of cells was observed by electron microscope, and apoptosis was verified by a flow cytometer. Results After treated with costimulate activation of PBMC with CD28 and CpG ODN, the inhibition rate of SW 480 cells was 68.35 %. When these two reagents were used in combination, the inhibition rate was 96.31%. By MTT method, combination activated PBMC and CI-988 possessed superior killing effect in comparison to anyone of them ( P 〈 0.01 ). Electron microscope and flow cytometer verified that cells were apoptosized after exposed to activated of PBMC with CI - 988 at 72 h.Conclusion Gastrin receptor antagonist can elevate the killing effect of anti - CD28/CpG ODN stimulated cells on colorectal carcinoma cell lines in vitro in two ways:necrosis and apoptosis.
