摘要
牛血红细胞铜锌超氧物歧化酶(Cu·Zn-SOD),经溶血,CuCl2处理及离子交换柱层析等步骤被纯化到高度均一程度.纯化酶在聚丙烯酰胺梯度凝胶图谱上的活性染色带与蛋白染色带相对应,在SDS-PAGE图谱上呈现单一区带.元素分析表明该酶含有2个锌原子和多于2个的铜原子.分子量和亚基分子量分别为33500和17800.在紫外区酶有4个明显吸收峰,分别为274.6nm,266.2nm,258.8nm和245.4nm.该酶对热稳定,纯化后酶比活性为14525u/mg,活力回收率为45%.本文对经CuCl2处理与氯仿-乙醇处理纯化的牛血Cu·Zn-SOD进行比较,结果表明。
Bovine erythrocytes Cu·Zn-SOD was purified by hemolysating,treatment with CuCl 2 and DEAE-Cellulose ino exchange chromatography.On gel concentration gradient electrophoresis chromatogram,the stainde bands of the enzymic activity corresponded with that of enzymic protein.The enzyme contained 2 atons of Zn and more than 2 atoms of Cu.Its M.W.and its S.M.W.were 33500 and 17000 respectively.The enzyme exhibited fourth absorption peaks in U.V.at 274.6nm,266.2nm,258.8nm and 245.4nm,and was stalbe under the condition of 70℃.The purified enzyme was 14525 U/mg,the rote of recovery of activity was 45%. In this paper,we made a comparison between some properties of SOD from treatment with Alc-CHCl 3 and CuCl 2.The results show that the treatment of Alc-CHCl 3 influenced some properties of enzyme.
