摘要
参照McCord＆Fridovich法,从绵羊血中提取纯化了超氧化物歧化酶,每2L血可获得纯化酶29.2mg,比活为12730U/mg,产率约63.4%.绵羊血超氧化物歧化酶呈淡蓝绿色,分子量33000,在276nm处有一个特殊的紫外吸收峰,在60℃以下稳定,pH6～11不影响其活性.酶液在聚丙烯酰胺凝胶盘状电泳上呈现1条带.
Superoxide dismutase (SOD) is an enzyme which catalyzes the dismutation of su-peroxide radicals. The authors extracted and purified SOD from sheep erythrocytes with chloro-form-ethanol treatment, K2HPO4 extraction , acetone precipitation and chromatography on DEAE-Sephadex A-50. 29. 2 mg purified enzyme can be obtained from 2000 mL sheep blood. Its specific activity is 12730 U/mg. The yield is 63. 4%. Sheep erythrocyte SOD is pale blue-green in color. It's molecular weight is 33000. There is a special ultraviolet absorption spectrum at 276 nm. It is stable below 60℃ and at pH 6-10. It reveals a band in polyacrylamide gel elec-trophoresis.
