Purification and Characterization of Copper/Zinc Superoxide Dismutase from Bovine Erythrocytes

摘要 牛血红细胞铜锌超氧物歧化酶经氯仿-乙醇混合液处理,丙酮沉淀,凝胶过滤和DEAE-纤维素柱层析等步骤提取和纯化.纯化酶在聚丙烯酰胺凝胶电脉(PAGE)图谱上的两条蛋白染色带和活性染色带相互对应,在SDS—PAGE聚丙烯酰胺凝胶电泳图谱上呈现单一区单一区带,说明该酶已纯化到均一程度.此酶分子量为34000,亚基分予量为17000,在紫外区最大吸收值为258nm.该酶对KCN和H2O2均敏感,在0~75℃温度范围内对热稳定.纯化酶的比活性为7463U/mg,纯化1009倍,酶的回收率为79%. A very yield of Cu. Zn-SOD from bovine erythrocytes has been obtained by treatment with Alc-CHCl3, preciptation with acetone, Sephadex G- 100 and DEAE -cellulose column chromatography.The purified SOD was shown to be highly homogeneous by analysis of PAGE and SDSPAGE. Its M. W. and its subunit M. W. were 34000 and 17000 respectively. The enzyme exhibited an unique UV-spectrum with a maximun at 258urn. The enzyme were inacted by KCN and H2O2 respectivly, and at least up to 75℃ has stabilily.The enzyme's specific activity was 7463 units/mg protein and was purified 1009 fold,the recovery of total activity was 79%.
