摘要
目的通过荧光金逆行标记的方法,检测腺病毒(adenovirus,Ad)介导的睫状神经营养因子(ciliary neurotrophic factor,CNTF)对视神经钳夹伤大鼠视网膜神经节细胞(retinal ganglion cell,RGC)数目的影响。方法采用钳夹视神经法制作大鼠视神经损伤模型,夹伤后向大鼠伤眼内注射Ad-CNTF,在取材前7d进行荧光金逆行标记RGC,于伤后28d,对大鼠的视网膜铺片进行荧光金标记的RGC记数。结果伤后28d,单纯损伤组、PBS组、Ad-LacZ组视网膜标记RGC数均较低;CNTF处理组视网膜标记RGC数为每视野(12.11±2.29)个(n=9),高于前3组,且差异非常显著(P<0.01)。Ad-CNTF组视网膜标记RGC数为每视野(20.93±2.67)个(n=8),在各组标记RGC数中最多,也好于CNTF组,2组间差异非常显著(P<0.01)。结论视神经损伤后眼内单次注射Ad-CNTF,可明显增加钳夹伤后28d大鼠视网膜的标记RGC数。与单次注射CNTF相比,对损伤的RGC具有更加显著的神经保护作用。
Objective To investigate the numbers of retinal ganglion cells retrogradely labeled by fluorogold after adenovirus delivered ciliary neurotrophic factor(CNTF) into eyeballs of the optic nerve injured rats. Methods Adenovirus with CNTF gene was injected into vitreous of the eyeballs of optic nerve crushed model rots. Retinal ganglion cells were retrogradely labeled by fluorogold 7 days before getting the retinas of rats. The numbers of retinal ganglion cells retrogradely labeled by fluorogold were investigated 28 days after crushed. Results The numbers of labeled retinal ganglion cells of control group,PBS group and Ad-LacZ group were lower than other groups 28 days after crushed; the numbers of labeled retinal ganglion cells of CNTF group were (12.11±2.29) per field (n = 9) and higher than three groups from above, there had very significantly different ( P〈0.01 ). The numbers of labeled retinal ganglion cells of Ad-CNTF group were (20.93±2.67) per field ( n = 8 ), were higher than other groups, and the values between Ad-CNTF group and CNTF group had very significant difference (P〈 0.01 ). Conclusion CNTF delivered by adenovirus into eyeballs of rats after optic nerve injury increased the numbers retinal ganglion cells of retrogradely labeled by fluorogold. The neuroprotected effects of Ad-CNTF are better than CNTF.
