高效凝胶渗透色谱法测定双灵固本散中多糖肽的分子量及其分布 被引量:6

Determination of Molecular Weight and Molecular Weight Distribution of Polysaccharide Peptides in Sun recomel(CGLE) by HPGPC

摘要 目的建立双灵固本散分子量及其分布测定的质控方法。方法采用高效凝胶渗透色谱法(HPGPC),多糖专用凝胶柱TSKG3000SWXL。以已知分子量右旋糖酐(Dextran)为对照品,0.2mol·L^-1硫酸钠溶液为流动相;柱温35℃;流速0.5ml·min^-1示差折光检测嚣。结果利用GPC专用软件处理测得双灵固本散中多糖肽的重均分子量(Mw)在12000-18000和数均分子量(Mn)在6500-7500以及分子量分布(Mw/Mn)小于3。用双灵固本散高效凝胶渗透图谱与灵芝子实体中分离出具有活性成分的多糖肽(GL—PPT)图谱作对照,结果表明,双灵固本散含有具有药理活性物质——灵芝多糖肽基本相同的分子量及其分布。结论本文实验建立的方法简便、快速,可作为灵芝糖肽的特征鉴别图谱和产品的质量控制。 OBJECTIVE A HPGPC method was developed to determine the molecular weight (Mw) and molecular weight distribution (MWD) of polysaccharide peptides in sun recome (CGLE--PPT), which can be used for quality control of CGLE. METHODS CGLE--PPT was analyzed with a special gel column for polysaccharides (TSKG3000SWXL) and a differential refractive index detector (RI detector), using dextran with known molecular weight as reference substance, and 0.2 mol · L^-1 sodium sulfate as mobile phase. The column temperature was 35C, and the flow rate was 0.5 ml · min^-1. RESULT The obtained data were calculated with special software for gel permeation chromatography (GPC). The Weight- average Molecular Weight (Mw) and the Number-average Molecular weight (Mn) of CGLE--PPT was 12000-18000 and 6500-7500 respectively, with the ratio of Mw/Mn (molecular weight distribution) being less than 3. The comparison of the chromatogram of CGLE--PPT and that of active polysaccharide peptides extracted from Ganoderma lucidum fruit body (GL-PPT), showed that CGLE has pharmacological active polysaccharide peptides which have almost the same Mw and MWD as those of GL--PPT. CONCLUSION The method is simple, convenient and rapid. It provides a characteristic chromatogram for identification of Ganoderma lucidum polysaccharide peptides and can be used for quality control of CGLE.
