摘要
选用NaOH浓度、菌液浓度、CaCl2浓度和搅拌提取时间为实验因素,每个因素拟定3个水平,运用L9(34)正交表,通过正交实验确定酵母硒核酸的最适提取条件。方差分析法处理实验结果后表明:15%酵母浓度、60min搅拌时间、1.0%NaOH和2.0%CaCl2是提取酵母硒核酸的适宜条件。用电感耦合等离子发射光谱法对硒核酸中硒含量进行了测定。提取的硒核酸中硒的含量为4257.207μg/g。
To study the suitable extraction methods of RNA from selenium-yeast by utilizing orthogonal experimental design, Four factors (concentration of yeast, the concentration of alikali, the concentration of calcium chloride and the reaction time) were researched. The results of orthogonal experiment indicate that the yeast of 15%, the alikali of 1.0%, the calcium chloride of 2.0%, and the reaction time of 60 minutes were the best suitable condition for the isolation of yeast RNA. The content of the selenium drawn in the nucleic acid is 4257.207μg/g by ICP-AES.
