摘要
我们以反相高效液相色谱法(RP-HPLC)和放射免疫测定法(RIA)研究了粉防己碱(Tet)对体外用钙离子载体A_(23187)诱导兔多形核白细胞(PMN)合成白三烯B_4(LTB_4)和血栓素A_2(TXA_2)的影响,并以地塞米松(Dex)和氟奋乃静(FPZ)作对照。结果表明,Tet与PMN温育15min即可抑制PMN合成LTB_4和TXA_2,其效应呈剂量依赖性。加入外源性花生四烯酸(AA)时,Tet的上述抑制作用消失。FPZ与Tet有相似的作用,Dex则与Tet不同,它需与PMN温育60min才出现抑制作用。
The effects of tetrandrine (Tet) on the production of leukotrine B4(LTB4) and thromboxaneB2(TXB2)of calcium ionophore A23187-stimulated rabbit polymorphonuclear leukocytes (PMN)are measured with the reversed phase high performance liquid chromatography and radioimmunoassay respectively. Dexa-methasone (Dex) and fluphenazine (EPZ)are used as the control. The production of LTB4 and TXB2 by PMN is significantly depressed by Tet in a dose-dependent mannet when incubated for 15 min. The IC50 for inhibition of LTB4 and TXB2 production is 8.3 and 9.6 μmol/L respectively. The characteristics of the effect of Tet are the same as those of FPZ.The effect of Dex occurs when incubated with PMN for 60 min.
