IL-1α对体外培养人牙囊细胞TNF-α表达的影响 被引量:3

摘要 目的:研究白细胞介素-1(αinterleukin-1α,IL-1α)对体外培养的人牙囊细胞(dentalfolliclecells,DFC)分泌肿瘤坏死因子-(αtumornecrosisfactor-α,TNF-α)的影响,探讨TNF-α在牙齿萌出过程中的作用。方法:原代培养人牙囊细胞;取3~6代细胞,培养基中加入IL-1α,在不同浓度和时间点,MTT检测细胞增殖,免疫组化法检测细胞中TNF-α的表达,SandwichELISA测定培养上清中TNF-α的含量。结果:IL-1α在25ng/ml,促进人牙囊细胞的增殖,IL-1α与牙囊细胞共孵育,可使TNF-α表达和分泌增强。结论:IL-1α有增强人牙囊细胞TNF-α表达的作用。 Objective:To study the effects of interleukin-1α (IL-1α) on the secretion of tumor necrosis factor-α (TNF-α) in human dental follicle cells. Method: The culture of human dental follicle cells was established, and the third to sixth passage cells were incubation with IL-1α for both time and dose dependent studies. MTT assays were used to evaluate the cells proliferation ; enzyme linked immunosorbent assays were used to measure the amount of TNF-α in the conditioned mediums and immunohistochemical staining technique was used to observe the expression of TNF-α. Result: This study showed that proliferation activity and the secretion of TNF-α increased significantly after incubation of the human dental follicle cells with 25 ng/ml IL-1α. Conclusion: IL-1α can increase the expression of TNF-α of human dental follicle cells at some stage during tooth eruption.
