矿化液对体外培养的人牙囊细胞碱性磷酸酶活性的影响 被引量:4

摘要 目的:探讨矿化液对体外培养的人牙囊细胞中碱性磷酸酶表达的影响。方法:取12~13岁患者因正畸原因拔除的下颌第三磨牙牙囊,原代培养人牙囊细胞,将第5代细胞加入矿化液培养,应用碱性磷酸酶组织化学方法染色、碱性磷酸酶试剂盒检测人牙囊细胞中碱性磷酸酶的活性。结果:培养的牙囊细胞呈长梭形、不规则多角形。免疫细胞化学染色显示抗波形丝蛋白染色阳性。矿化液诱导7d,碱性磷酸酶染色呈阳性,碱性磷酸酶活性检测第3、5、7d明显升高,与对照组均有显著性差异。结论:矿化液能增强体外培养的人牙囊细胞中碱性磷酸酶的表达。 AIM:To evaluate the effect of mineralized condition on alkaline phosphatase (ALP) activity of human dental follicle cells ( DFCs ) in vitro. METHODS : Human DFCs of the fifth passage were cultured in mineralized condition( 10 nmol/L dexamethasone,50 mg/L vitamin C, 10 mmol/L β - sodium glycerophosphate and 50 mL/L FBS in DMEM) ,while the control cells were cultured in DMEM with 5% FBS. The human DFCs were observed with inverted microscope. The alkaline phosphatase level of human DFCs was investigated with histochemical method and ALP assay kit. RESULTS:The cultured human DFCs in vitro showed fibroblastic morphology and polygon. Positive expression of vimentin was identified by immunohistochemistry staining. ALP activity level of experimental group was greatly increased following 3,5,7 d inducement by mineralized condition,and was significantly different from that of the control. Positive staining of ALP was demonstrated after 7 days inducement. CONCLUSION:ALP activity of human dental follicle cells could be enhanced in the mineralized condition in vitro.
