人骨骼肌失神经后胶原纤维和DNA的定量变化 被引量:3

CHANGE OF CELL PROLIFERATION AFTER HUMAN SKELETAL MUSCLE DENERVATION:AQUANTITATIVE COLLAGEN AND DNA STUDY

摘要 目的:了解人骨骼肌失神经后细胞增殖状态的变化规律,对反映肌肉不可逆改变的指标进行初步探讨。方法:42例周围神经损伤患者术中活检。①HE染色测肌细胞的截面积,②VanGieson染色测定胶原和肌细胞面积比,③CAS200图像分析系统DNA定量测定方法统计S期细胞核比例。结果:神经损伤后不同时间肌细胞的截面积无明显差异;胶原纤维与肌细胞面积比随失神经时间延长明显增高;S期细胞核所占百分比的变化以失神经3~4个月时最高,以后减少。结论:人骨骼肌细胞失神经支配后3至4个月内保持高增殖水平,以后降低,故神经修复术应力争早期。细胞增殖状态指标是反映肌萎缩是否可逆的潜在指标,可能有助判断神经修复手术的预后,指导临床对手术方法的选择。 Objective:To investigate the changes of cell proliferation after skeletal muscle denervation in human,Methods:Biopsies were done in 42 cases of peripheral nerve injuries,The cross areas of muscle cells were measured by VIDS Ⅲ Image Analysis System in HE stained sections.The percentages of S phase cell nucleus were calculated using Image Cytometry/DNA quantitative analysis with CAS 200 System,Results:No statistical difference was found between different time groups after nerve injuries in cross sectional area of muscle cells. Collagen/muscle cell ratio of area increased significantly as denervation prolonged.The percentage of S phase nucleus was high at 3~4 months and then decreased.Conclusion:The proliferation of human skeletal muscle cell remains at high level 3~4months after denervation and then slows down.This suggests that nerve repair should be performed early.Examination of cell proliferation is possible to indicate if the denervation of cells is irreversible.So it is helpful in choosing proper surgery and prognosing the effect of nerve repair.
