双液相萃取对亚麻籽分离蛋白功能特性的影响 被引量:7

摘要 采用含水乙醇正己烷双液相(TPS)和正己烷单相(SP)萃取的亚麻籽粕为研究对象,通过碱溶酸沉提取分离蛋白,应用质构仪和差示扫描量热仪等研究分离蛋白的功能特性.结果表明:双液相技术能明显改善亚麻分离蛋白的功能特性,TPS分离蛋白的凝胶力学性质得到明显增强;TPS分离蛋白的保水性、保油性、起泡能力和起泡稳定性优于SP分离蛋白,而SP分离蛋白的溶解性和乳化能力略优于TPS分离蛋白;双液相萃取有利于提高分离蛋白的变性温度,SP分离蛋白的变性温度与TPS分离蛋白的变性温度分别为99.7℃与108℃. Two types of protein isolates were prepared from flaxseed (Linum usitatissimum) defatted flour extracted by aqueous ethanol/hexane two-phase-system (TPS) and hexane single-phase (SP) respectively, with means of alkaline extraction and acid precipitation of proteins at the isoelectric point. Main functional properties of protein isolates were determined. The gel mechanical properties, fat absorption capacity, water absorption capacity, emulsion stability, foaming capacity and foaming stability of TPS protein isolates were higher than that of SP protein isolates except for solubility and emulsion capacity. Thermal properties Of protein isolates were also investigated by differential scanning calorimetry (DSC). A significant denaturation temperature ( Td = 108℃ ) of TPS protein isolates is higher than that of SP protein isolates ( Td = 99.7 ℃ ).
