Exploration on the activated state of RAS/MAPK signaling pathway in esophageal cancer and its clinical significance

摘要 目的通过研究细胞外信号调节激酶(ERK)在食管癌组织、癌旁组织、正常组织中的表达和激活情况,探讨RAS/MAPK信号级联在食管癌发生中的意义。方法Western-blot方法检测15例新鲜食管癌组织及相应的13例癌旁组织、14例正常组织中p-ERK、ERK的表达。免疫组化方法检测该批组织石蜡切片中p-ERK的表达及其细胞定位。结果Western-blot显示全部组织均表达ERK和p-ERK,统计分析表明,ERK在3种组织中的表达差异无统计学意义(P>0.05);p-ERK在正常组织、癌旁组织、癌组织中的表达有下降趋势,其在癌组织中的表达显著低于正常组织(P<0.05)。免疫组化则显示全部组织均表达p-ERK,阳性信号主要位于食管黏膜上皮细胞和癌细胞的胞核。结论RAS/MAPK信号转导通路的活化存在于所有食管癌组织、癌旁组织和正常组织,食管癌中该通路的活化水平显著低于正常组织,该通路活化水平的改变可能与食管癌的发生有关。 Objective To investigate the possible function of RAS/MAPK signaling pathway in the occurrence of esophageal cancer by studying, the expression and activation of ERK in esophageal cancer, the corresponding adjacent tissues and histologically normal tissues. Methods The expression of p-ERK and ERK in 15 fresh esophageal cancers, the corresponding 13 adjacent tissues and 14 histologically normal tis.sues were tested by Western-blot. Meanwhile the expression and distribution of p- ERK in the above mentioned tissues were tested by immunohistochemistry method, Results Westernblot showed that ERK and p-ERK were expressed in all the tissues tested. The expression of ERK .showed no significant difference in the three kinds of tissues (P〉0.05). The expression of p-ERK in the histologically normal tissues, adjacent tissues and cancer showed a decrease trend. Furthermore, its expression in cancer was significantly lower than that in histologically normal tissues (P〈0.05). Immunohistochemistry showed that p-ERK was expressed in all the tissues tested in which positive signals were detected mainly in the nuclei of the esophageal epithelial cells and cancer cells. Conehtsion The signaling pathway of RAS/MAPK is activated in all the esophageal cancer, adjacent tissues and histologically normal tissues; the activation of this pathway in cancer is significantly lower than that in histologically normal tissues. Therefore the change of this signaling pathway's activated state may be involved in the occurrence of esophageal cancer.
