健脾化瘀方对胃癌细胞SGC-7901凋亡和周期的影响 被引量:3

摘要 目的:观察健脾化瘀方体外对人胃癌细胞株SGC-7901增殖、凋亡及细胞周期的影响。方法:①采用MTT法观察不同浓度健脾化瘀方体外对人胃癌细胞株SGC-7901增殖的抑制作用;②用Annexin V/PI荧光双染法检测健脾化瘀方诱导肿瘤细胞进入凋亡的影响;③用流式细胞仪检测健脾化瘀方对SGC-7901细胞周期的阻滞效应。结果:①健脾化瘀方能抑制人胃癌细胞SGC-7901增殖,随着药物浓度的增大以及作用时间的延长,其对细胞的抑制率也明显增强,呈现剂量及时间依赖性。②当2mg/ml和4mg/ml健脾化瘀方作用于人胃癌SGC-7901细胞48h可显著诱导细胞凋亡的产生。③细胞周期方面,健脾化瘀方作用于人胃癌细胞SGC-7901后G2/M期比例与阴性对照组相较显著上升,G0/G1期稍有上升,而S期比例下降,表明细胞被阻滞于G2/M期。结论:健脾化瘀方可明显抑制人胃癌细胞SGC-7901的增殖作用并能够诱导该细胞凋亡,改变细胞周期。 Objective: Observing the effect of Jianpi Huayu Recipe on Inducing Cell Apoptosis and Cell Cycle of Human Gastric Carcinoma Cell Line SGC-7901.Methods: After SGC-7901 cells were treated with Jianpi Huayu Recipe at different concentrations, cell proliferation was observed by MTT assay.The effect of carcinoma cell apoptosis were detected by Annexin V/PI and the retarding effect of cell cycle were determined by flow cytometry(FCM).Results: ①Compared with the negative control group,Jianpi Huayu Recipe significantly inhibited the proliferation of SGC-7901 cells.The inhibition effect increased with the dose of Jianpi Huayu Recipe and response time;②When the cells were treated by 2mg /ml and 4mg /ml of Jianpi Huayu Recipe, cell apoptosis was detected by cytometer;③As compared with the negative control group,when the cells were treated by Jianpi Huayu Recipe,the percentage of the cells in G2 /M phase increased obviously in treatment group,while the percentage of the cells in G0 /G1 phase increased slightly,and S phase decreased,which showed the cells were arrested at the G2 phase of the cell cycle.Conclusion: Jianpi Huayu Recipe can inhibit the proliferation of human gastric carcinoma cell line SGC-7901 and can induce this cell apoptosis in vitro.
