摘要
采用水溶醇沉法从胖大海中提取水溶性粗多糖,通过棉纤维和DEAE-SephroseCL-6B离子交换柱分离得到含量较大的酸性多糖ASPⅠ;经凝胶色谱和高效凝胶渗透色谱鉴定其为均一组分,气相色谱和离子色谱测其单糖组成(质量比)为鼠李糖:阿拉伯糖:半乳糖:葡萄糖:木糖:半乳糖醛酸=24.55:14.22:10.45:1.84:1.22:28.05。红外光谱测定表明,ASPⅠ具有多糖的特征吸收峰。
Acidic polysaccharide ASP Ⅰ was obtained by ethanol depositing and using diethylaminoethyl cellulose and DEAE-Sephrose CL-6B column. It was identified homogeneous polysaccharide by gel filtration and high performance gel penetrate chromatography. It was composed of rhamnose, arabinose, galactose, glucose, xylose and galacturonic acid by gas chromatography and ion chromatography analysis, and their ratio was 24.55:14.22: 10.45:1.84:1.22:28.05. FT-IR showed ASP Ⅰ had characteristic absorption peaks of polysaccharide.
