摘要
以欧李3号、4号品种为试材,初步建立了茎尖组织培养的快繁体系。结果表明:欧李对激素反应比较敏感。分化基本培养基采用MS优于B5和W h ite。在MS+0.25mg.L-1BA+0.1mg.L-1IAA的分化培养基上,萌发速度快,成苗率高。增殖培养时,增殖系数随BA浓度增大而提高;欧李3号在MS+0.5mg.L-1BA+0.075mg.L-1NAA的培养基上,增殖系数为5.43;欧李4号在MS+0.4mg.L-1BA+0.05mg.L-1NAA的培养基上,增殖系数为4.56,增殖效果较好。壮苗培养时,欧李3号适宜的培养基为改良MS+0.075mg.L-1NAA,欧李4号适宜的培养基为改良MS+0.05mg.L-1NAA。生根培养时,欧李3号适宜的培养基为1/2MS+0.4mg.L-1NAA;生根率达72.3%,欧李4号适宜的培养基为1/2MS+0.2mg.L-1NAA;生根率达74.6%。移栽采用草炭、田园土、珍珠岩(5∶2∶1),成活率达72.7%。
Rapid propagation of shoot -tip culture system was set up with Ouli3 and 4 as materials. The main results were as follows: Ouli was found sensitive to hormone. Medium MS was better for differentiation than B5 and white. The high rate of bourgeon and survival occurred in Medium MS + 0. 25 mg · L^-1 BA + 0. 1 mg· L^-1 IAA. As for multiplication culture, positive correlation was observed with multiplication coefficient and BA concentration. The coefficients were 5.43 and 4. 56 for Ouli 3 in MS +0. 05 mg· L^-1 BA +0. 075 mg· L^-1 NAA and for Ouli 4 in MS +0. 4mg· L^-1 BA + 0. 05 mg · L^-1 NAA, respectively. The modified medium MS +0.75 mg· L^-1 NAA was optimum for healthy seedling culture of Ouli 3, and the best one for Ouli 4 was MS +0. 05 mg · L^-1 NAA. 1/2MS +0.4mg· L^-1. NAA was the optimum medium for rooting culture of Ouli 3, while for Ouli 4,the optimum one was 1/2MS + 0. 2mg· L^-1 NAA. The rooting rate was 72.3% and 74. 6% respectively. Survival rate was 72.7% when the seedlings were transplanted in peat + garden soil + peadite(5: 2:1 ).
