快速准确制备大肠杆菌高效感受态细胞中有关OD_(600)值的探讨 被引量:8

摘要 目的探讨在不同转速下大肠杆菌OD600值与时间的关系,为能快速而准确的制备高效感受态细胞提供OD600值的量化标准。方法在不同转速下每隔一定时间测定大肠杆菌OD600值。结果总结了不同转速下OD600值在0.3—0.4所需时间段,并且用此时间段培养的细菌制备的感受态细胞的转化效率可高达(6.45±0.05)×107转化子/μg DNA。结论用此法制备的感受态细胞完全可以满足大部分在质粒中进行的常规克隆的需要。 Objective To investigate the relationship between the OD600 of E. coli and the time in different rotation speed and obtain the quantization standards of the OD600 of E. coli. Methods The OD600 of E. coli was determined every few minutes in different rotation speed. Results It showed that the periods of the OD600 (0.3~0.4 ) in different rotation speed, according to the tests, The transformation efficiency of this method reaches to (6.45 ± 0.05) × 107 cfu/μg DNA. Conclusion It could completely meet the most requirements of all kinds of transformation experiments with this method.
