巨柏细胞色素氧化酶同工酶变异分析 被引量:9

The isoenzymic variation analysis of the cytochrome oxidase of the Cupressus gigantea

摘要 为阐明巨柏Cupressus gigan tea的濒危机理,根据聚丙烯酰胺琼脂电泳对巨柏3个大居群的36个个体功能叶片的细胞色素氧化酶的同工酶谱带测定结果进行编码,对各酶谱带进行聚类分析;同时结合细胞色素氧化酶的3个遗传指标进行分析。结果表明,来自同一居群的个体,同工酶谱带相似性高,遗传杂合度小(HeA=0.726,HeB=0.691,HeC=0.704),相似性系数大,遗传距离小,这说明居群内个体遗传分化小;而不同居群间的个体则恰好相反(IAB=0.240,IAC=0.259,IBC=0.401;DAB=1.427,DAC=1.351,DBC=0.914),这主要是由青藏高原特殊的气候条件和巨柏的地理分布格局所造成的。 To illustrate the endangerous mechanism of Cupressus gigantea, the cytochrome oxidase (CYT)isoenzymic bands of leaves of 36 plants from 3 isolated population were determined by polyacrylamide gel electrophoresis. Meanwhile the cluster analysis of the isoenzymic bands has been made and 3 genetic indexes of CYT were analysed. The results indicate that individuals from the same population show higher similarity in isoenzymic bands and genetic heterozygosity is small( HeA = 0. 726, HeB = 0. 691, HeC = 0. 704), genetic identity is big, genetic distance is small, which illustrate genetic variation of inner population is very small. However,the results of the individuals from different population is contrary to that from the same population, that is, genetic identity is small and genetic distance is big ( IAB = 0.240,IAB=0.259, IBC =0.401; DAB=1.427, DAC=1.351, DBC=0.914). The main reason is the special climate condition of Qinghai-Tibet Plateau and geographic distribution situation of Cupressus gigantea.
