摘要
目的 探讨细胞因子诱导的杀伤细胞(CIK)的生物学特性及抗肿瘤作用。方法 用淋巴细胞分离液分离健康献血者血中单个核细胞,用IFN-γ、IL-1β、IL-2、CD3单抗诱导CIK细胞。以CD3AK细胞作为对照。流式细胞仪检测细胞表型;乳酸脱氢酶(LDH)释放法分析细胞毒活性。结果 CIK细胞和CD3AK相比,在前期细胞增殖能力无明显差异,但至第10~21天时CIK细胞的增殖能力显著高于CD3AK细胞(P〈0.05)。诱导14d时,CD3^+细胞和CD3^+CD56^+阳性细胞显著增多(分别约占84.75%和33.45%)。不同效靶比例的CIK细胞对K562细胞、BeL-7402细胞和Hela3细胞的溶瘤率均显著高于CD3AK细胞(P〈0.01),在40:1的效靶比时,其杀瘤活性最强。但CIK细胞对不同肿瘤细胞的杀伤率比较无显著性差异(P〉0.05)。结论 CIK细胞是CD3^+CD56^+双阳性细胞群,其增殖能力及杀瘤活性明显优于CD3AK细胞。CIK细胞可作为一种新的广谱、高效的免疫效应细胞应用于临床。
Objective To investigate the biological characteristics of cytokine induced killer (CIK) cells and its anti-tumor activity in vitro, Methods Peripheral blood monoclear cells from healthy donors were induced into CIK cells with IFN-γ, IL-1β, IL-2 and anti- CD3 antibody. CD3AK (anti CD3 Monoclonal antibody activated killer ceils) were prepared as a control. The cellular phenotype were detected by FCM and the cytotoxicity was measured by LDH release assay. Results After 10 days induction, the proliferation rate of CIK cells was markedly higher than that of CD3AK (P 〈 0.01 ). After 14 days' induction, CIK cells with CD3^+ population was about 84, 75% ,and the proportion of CD3^+ CD56^+ NKT subset cells was about 33.45%, CIK cells showed higher lysis rates to K562,BeL-7402 and Hela3 tumor cell lines than those of CD3AK cells at differeht effector to target ratio (P 〈 0.01 ), Conclusion CIK ceils have higher proliferation potency and potent cytotoxicity on tumor cells than CD3AK cells in vitro, CIK cells would be a new effect cells to be applicated in clinic.
