摘要 目的 比较DC对诱导4 d(L4) 和诱导7 d(L7) 的LPAK 细胞体外杀伤人肝癌细胞株BEL- 7402(B) 的杀伤力和杀伤模式的影响.方法 L4 组为B+ L4 ,LD4 组为L4 组+ DC;L7 组为B+ L7 ,LD7 组为L7 组+ DC.L4 和L7 与B的效靶比均为5∶1 和10∶1两种. 采用杀伤细胞检测技术及电镜技术,比较各组的杀伤效应和杀伤模式.结果 各组的细胞毒活性为LD4 > L4( P< 0-01) ,LD7 > L7( P< 0-01) .L4 组和LD4 组的BEL7402 细胞呈现不同程度的变性坏死,L7 组和LD7 组的BEL7402 细胞则呈现不同程度的凋亡改变.结论 DC对不同诱导时间的LPAK 细胞体外杀伤肿瘤细胞都有明显的促进作用,但并不改变LPAK 细胞对肿瘤细胞的杀伤模式. AIM To study the efficiency of dendritic cells (DC) on LPAK cells induced for 4 days (L 4) and 7 days (L 7) in killing human hepatoma cell line BEL 7402(B) in vitro, including killing activity and model. METHODS Two kinds of ratios of effect and target (5∶1 and 10∶1) were used in L4 group (B+L 4), LD4 group (B+L 4+DC), L7 group (B+L 7) and LD7 group (B+L-7+DC). The techniques of cell killing detection and electronic microscopy were adopted. RESULTS The cytotoxic activities were LD4>L4 ( P <0 01),LD7>L7 ( P <0 01). The BEL 7402 cells in L4 and LD4 groups revealed denaturation and necrosis in each degree, but the BEL 7402 cells in L7 and LD7 groups manifested apoptotic change in a certain degree. CONCLUSION DC could enhance the killing activity of LPAK cells induced at different times against tumor cells in vitro, but they could not change the killing model of LPAK cells.
