摘要
目的探讨中药保心合剂对扩张型心肌病免疫作用的机制。方法采用柯萨奇B3病毒(CVB3)致慢性自身免疫损伤心肌病模型,用保心合剂进行治疗,采用免疫转印技术检测小鼠外周血清中抗ADP/ATP载体抗体、抗β1受体抗体,采用ELISA技术检测小鼠外周血清中肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的含量。结果模型组心肌结构显示散在纤维化,抗ADP/ATP载体抗体、抗β1受体抗体检出率分别为77.78%、100.00%。给药组TNF-α及IL-6含量较空白模型组下降明显(P<0.05),3种剂量给药组之间无统计学意义。结论保心合剂对扩张型心肌病具有免疫调节及心肌保护作用。
Objective To probe the protective role of Baoxin Mixture on chronic autoimmune myocardium lesion induced by CVB3 virus. Methods The chronic autoimmune myocardium lesion models were prepared with CVB3 virus. The change of the mice microscopic structure were observe and the percentage of the autoantibody against myocardiurn mitochondria ADP/ATP carrier and the autoantibody against β- agrenoceptor and the level of TNF - α and IL - 6 were tested by ELISA kits. Results (1)The microscopic structure:Under the optical microscope, the myocardial injury in normal group was characterized by low scores of fibrosis associated with calcification lesions. The changes of the 3 drug groups were similar, only a few of calcifications remained. (2)The autoantibody A against myocardium mitochondria ADP/ATP cartier and the autoantibody against β - agrenoceptor: The percentage of Gegenshu against myocardium mitochondria ADP/ATP carrier and the autoantibody against β- agrenoceptor in the model group were higher than that in the 3 drug groups(P〈0.05) ,There were no significant differences in the 3 drug groups( P 〉0.05 ). (3)The concentration of cytokines (tumor necrosis factor- A and interleukin- 6) :The level of cytokines was higher in the model group than in than that in the 3 drug groups and the normal group(P〈0.05 ). There were no significant differences between the 3 drug groups ( P 〉 0.05 ). Conclusion Baoxin Mixture had the function of immune regulation and myocardium protection. Its possible mechanism might relate to reduce autoantibodies against cardiac peptides by immune regulation, and reduce cytokines TNF-α and IL- 6.
