摘要
目的:建立洛美沙星人血浆及尿药浓度HPLC柱切换测定法。方法:LiChropredRP2(30mm×4mm,25~40μm)预处理柱,水为预处理流动相。ShimpackCLC-ODS分析柱(150mm×6mm,5μm),分析流动相为甲醇-0.2mol/L醋酸铵(pH2.0)(3268测血浆,3466测尿),紫外检测波长290nm。结果:洛美沙星血浆及尿最低检测浓度分别为0.1mg/L及5mg/L,线性范围0.125~6mg/L及50~1000mg/L,平均净化回收率74.2%±2.29%及893%±1.52%。结论:用于测定8名健康志愿者口服洛美沙星制剂400mg后血浆及尿药浓度。
OBJECTIVE:To establish a simple HPLC column switching method for measuring lomefloxacin in human plasma and urine.METHOD:Diluted plasma and urine were directly injected into the pretreatment column of LiChropred RP 2(30 mm×4 mm,25~40 μm),and were cleaned up by water.The Shimpack CLC ODS(150 mm×6 mm,5 μm) was used as analytical column.The mobile phase consisted of methanol 0.2 mol/L ammomiun acetate buffer(pH 2.0)(32∶68 for plasma,34∶66 for urine).The eluent was monitored at 290 nm.RESULTS:The limit of detection was 0.1 mg/L and 5 mg/L,linear range was 0.125~6 mg/L and 50~1 000 mg/L,the recovery of clean up was 74.2%±2.29% and 89.3%±1.52%,for lomefloxacin in human plasma and urine respectivly.CONCLUSION:This method was applied to the determination of lomefloxacin in eight Chinese healthy volunteers after a single oral dose of 400 mg.
