摘要
为研究仔猪肠道组胺免疫组织化学固定液、染色条件及组胺在仔猪空肠的分布情况,将5头三元杂交仔猪屠宰后,取其空肠,Carnoy氏液或4%多聚甲醛磷酸缓冲液固定,进行抗原修复及一抗作用条件的免疫组织化学方法学研究。通过显微测微技术测定空肠各层组胺阳性细胞的分布。结果发现,Carnoy氏液是肠道组胺免疫组织化学的良好固定液;柠檬酸缓冲溶液(pH7.2)100℃抗原修复3 h及一抗4℃孵育12 h是空肠组胺免疫组化的良好条件;黏膜及黏膜下层组胺阳性细胞数极显著地高于浆膜(P<0.01)。
To investigate the histamine locate in the jejunum in piglets by immunohistochemical method. Jejuna were taken from 5 piglets. The fixative fluid, the method of antigen renovating and condition of antigen incubating were studied. The counts of histamine positive ceils were measured using microcheck grid in jejunum. Carnoy's was suited to fix the tissue. It is the condition of immunohistochemistry that citric acid buffer fluid boiled for 3h to restore the antigen, and the antibody incubated with the section in 4℃for 12 h. The counts of histamine positive cells in mucosa and submucosa were significantly higher than that of in chorion. It is the good method of histamine immunohistochemistry that Carnoy's fixed the tissue, citric acid buffer fluid boiled for 3 h and antibody incubated with the section in 4℃ for 12 h. Histamine was abundant in jejunum of in piglets, especially in mucosa and submucosa.
