不明原因原发不孕症患者黄体中期血清蛋白质谱研究 被引量:2

Investigation of serum proteomic spectra in the patients suffering from primary infertility with unknown reasons during midluteal phase

摘要 目的探讨不明原因原发不孕症患者血清蛋白质谱特征,寻找女性不孕症血清生物学标志物。方法采用表面增强激光解离飞行时间质谱技术(SELDI-TOFMS),运用WCX2蛋白质芯片检测2005-01-11卫生部北京医院妇产科、北京大学第三临床医院生殖中心、北京妇产医院的29例不明原因原发不孕患者(原发不孕组)和30例健康育龄妇女(对照组)黄体中期血清蛋白质谱,建立并测试不明原因原发不孕症患者血清蛋白质指纹图谱模型。结果筛出差异显著的5种蛋白质组成不明原因原发不孕患者血清的蛋白质谱最佳优化模型,其中质荷比(m/z)分别为8690、8715、8634、8826的4种蛋白质表达下降,质荷比(m/z)6628的蛋白质表达上调。结论不明原因原发不孕患者与正常育龄妇女黄体期血清蛋白质谱存在明显差异,深入研究可能为女性不明原因不孕的病因学提供基础,同时有助于不孕症生物标志物的筛查和发现。 Objective To study the characteristics of the serum proteomic spectra in the patients suffering from primary infertility with unknown reasons in order to find the serum biologic markers of infertility. Methods Proteomic spectra of serum samples from 29 primary infertility patients with unknown reasons and 30 healthy women were analysis by SELDI- TOFMS (surface-enhanced laser desorption / ionization time of flight mass spectrometry). The patients and their control were taken from January to November of 2005 in the Department of Obstetrics and Gynecology of Beijing Hospital. The Reproductive Center of the Third Cilinical Hospital of Beijing University and Beijing Obstetrics and Gynecology Hospital. The peaks were detected by WCX2( weak cation exchange) proteinchip array first. A system of serum protein finger printing was developed for detecting primary infertility patients. Result Five proteins which were prominently different between two groups were obtained and they made up the markers pattern of infertility patients, with 4 protein (8690,8715, 8634,8826,m/z) expressions being down-regulated, but the 6628 m/z one was up-regulated. Conclusions There are prominent differences of serum proteomic spectra between the primary infertility patients with unknown reasons and healthy women in luteal phase. Further study may provide a base for the etiology of infertility women with unknown reasons and may be helpful for the discovery and screening of biologic markers for infertility women.
