摘要
目的:用高脂饲料与小剂量链脲佐菌素建立糖尿病大鼠模型,观察其骨的变化。方法:实验于2004-09/2005-12在广东医学院药理教研室、组胚教研室和骨生物研究室完成。取24只雌性SD大鼠单纯随机分成正常对照组(n=9),糖尿病组(n=15),正常对照组予普通饲料加生理盐水5mL/kg灌胃,糖尿病组予普通饲料加脂肪乳剂灌胃5mL/kg,1次/d。连续16周后,糖尿病组腹腔注射2g/L链脲佐菌素30mg/kg,继续用脂肪乳剂喂养6周;正常对照组腹腔注射等体积柠檬酸缓冲液,继续普通饲料喂养6周。实验结束后测体质量,取血测血糖和血脂,取胫骨做骨组织形态计量学分析。结果:18只大鼠进入结果分析。①糖尿病组大鼠高密度脂蛋白胆固醇浓度和体质量显著低于正常对照组,(P<0.05,0.01),血糖和三酰甘油显著高于正常对照组,(P<0.01,0.05)。②糖尿病组大鼠胫骨上段骨小梁面积百分数、数量、厚度及骨小梁荧光周长百分率明显低于正常对照组(P<0.05),骨小梁分离度和破骨细胞数目指数高于正常对照组(P<0.05)。结论:高脂饲料与链脲佐菌素建立的糖尿病大鼠模型合并有骨质疏松症。
AIM: To establish rat model of diabetes with intralipid and small-dose streptozotocin (STZ), and observe the changes of bones.
METHODS: The experiment was conducted in the Department of Pharmacology, Department of Histology and Embryology, Laboratory of Bone-Biomechanism, Guangdong Medical College between September 2004 and December 2005. Twenty-four femal SD rats were randomly divided into normal control group (n=9) and diabetes group (n=15). Rats in the normal control group received intragastric administration of 5 mL/kg normal saline and feedstuff, and rats in the diabetes group were treated with gastric perfusion of intralipid plus 5 mL/kg STZ once a day. After 16-week-administration, rats in the diabetes group were intraperitoneally injected with 2 g/L STZ 30 mg/kg, and then fed with intralipid for 6 weeks. Rats in the normal control group were intraperitoneally injected with samevolume citrate buffer solution, and then fed with normal feedstuff for 6 weeks. The levels of blood glucose (BG) and blood lipid as well as body mass were measured after the experiment, and the shin bone was analyzed with bone histomorphometry.
RESULTS: A total of 18 rats were involved in the analysis of results.① The concentration of high density lipoprotein cholesterol (C-HDL) and body mass in rats of diabetes group were significantly lower than those in the normal control group, (P 〈 0.05, 0.01 ), and the levels of BG and triglyeride (TG) were remarkably higher than those in the normal control group, (P 〈 0.01, 0.05).②The area percentage, quantity and thickness of bone trabecula in the superior segment of shin bone and percentage of fluorescent perimeter were remarkably lower than those in the normal control group (P 〈 0.05). The resolving power and index of osteoclast of bone trabecula were higher than those in the normal control group (P 〈 0.05).
CONCLUSION: Rat model of diabetes established by intralipid and STZ has osteoporosis.
