补肾益气类中药干预运动大鼠血生化学指标及抗氧化系统的变化 被引量:4

摘要 目的:观察补肾益气类中药对运动大鼠血生化学指标、血清超氧化物歧化酶活性、丙二醛水平的影响。方法:实验于2004-05/08在山东体育学院体育医院实验室完成。①选用Wistar大鼠80只,4月龄,雌雄各半。大鼠适应性喂养、游泳训练3d后,按随机抽签法分为2组:对照组和实验组,每组40只。实验组:灌胃含生药0.135g/mL补肾益气类中药胶囊溶解的混悬液10mL/(kg·d),该中药组方主要为鹿茸、红参、当归、白术(山东省中药药材公司),按比例配成胶囊剂型,含生药0.5g/粒,胶囊由山东中医药大学制剂室协助生产。对照组:灌胃等量生理盐水。两组均干预6周。②实验开始后2组大鼠即进行渐进性游泳训练。实验周期为6周,每次训练的时间以每周递增15min的方式。大鼠渐进性游泳训练6周后即进行尾部负重(加150g的重锤)的负荷力竭游泳实验,若出现沉在水下10s不能上浮和身体动作不协调时即为力竭,记录力竭时间。③分别于实验前,渐进性游泳训练2,4,6周后,负荷力竭游泳实验结束即刻于2组动物各取8只动物(雌雄各4只)。采用全自动细胞计数仪测定红细胞计数和血红蛋白含量。由采用光学显微镜计数网织红细胞。按购自南京建成生物技术公司试剂盒说明,采用比色法检测血清中超氧化物歧化酶活性和丙二醛水平。结果:大鼠40只均进入结果分析。①实验前,两组血红蛋白、红细胞计数、网织红细胞计数、血清超氧化物歧化酶活性和丙二醛水平差异不明显(P>0.05)。渐进性游泳训练2周后,血清超氧化物歧化酶活性明显高于对照组(P<0.05),丙二醛水平明显低于对照组(P<0.05)。渐进性游泳训练4,6周后和负荷力竭游泳结束后即刻,血清超氧化物歧化酶活性、网织细胞计数、红细胞计数、血红蛋白含量明显高于对照组(P<0.01),血清丙二醛水平明显低于对照组(P<0.01)。②实验组大鼠游泳力竭时间明显长于对照组[(15.32±2.56),(8.50±2.15)min,P<0.05]。结论:补肾益气类中药能明显改善运动大鼠血生化学指标,提高机体抗氧化能力,并且能显著增强大鼠耐力,延缓运动疲劳的出现。 AIM:To observe the effects of Bushen Yiqi Chinese herb on blood biochemical index, activity of serum superoxide dismutase (SOD) and level of malonaldehyde (MDA) in sport rats. METHODS:The experiment was conducted at the Laboratory of Physical Education Hospital, Shandong Institute of Physical Education and Sport from May to August 2004. ①A total of 80 Wistar rats aged 4 months, of either sex, were selected. Three days after rats were fed adaptively and received swimming training. They were randomly assigned into 2 groups: control group and experimental group with 40 rats in each group. The rats in the experimental group were treated with 10 mL/kg suspension dissolved Bushen Yiqi Chinese herb capsule containing 0.135 g/mL crude drug daily, which was mainly composed of pilose antler, red ginseng, angelica and atractylodes macreeephala (Shandong Provincial Chinese Medical Material Company) by gastric perfusion, and capsule was prepared in proportion, containing 0.5 g crude drug in each capsule that was produced in the help of Manufacturing Laboratory, Shandong University of Traditional Chinese Medicine. The rats in the control group were treated with saline of the same volume by gastric perfusion. The intervention was lasted for 6 weeks in the two groups. ②When the experiment was performed, the rats in the two groups received gradually swimming training for 6 weeks. The training duration increased by 15 minutes every time in each week. After gradually swimming training for 6 weeks, the rats were subjected to loading swimming training with tail weight leading (adding 150 g hammer). If the rats sunk in water for 10 s and could not float up as well as the action was not accordant, it was indicated that the rats were exhausted, and then exhausting time was recorded. ③Eight male and female (half and half) rats were selected from the two groups, respectively before experiment, 2, 4 and 6 weeks after gradually swimming training, immediately after loading swimming. Red blood cell count and content of haematogiobin were determined with full-antomatic cell events-per-unittime meter. Granulofilocytes were counted with light microscope. Serum activity of SOD and level of MDA were detected with chromatometry based on the instruction of kit of Nanjing Jiancheng Biotechnology Company. RESULTS: A total of 40 rats were involved in the result analysis. ①Before experiment, there were no significant difference in counting of haematogiobin, red blood cell and granulophilocytes, activity of SOD and level of MDA (P 〉 0.05). Two weeks after gradually swimming training, the activity of SOD in serum was significantly higher than that in the control group (P 〈 0.05). The level of MDA was markedly lower than that in thecontrol group (P 〈 0.05). Four and six weeks after gradually swimming training and immediately after loading swimming, the activity of SOD in serum, counting of reticulocyte and red blood cell, content of haematoglobin were obviously higher than those in the control group (P 〈 0.01 ), while the level of MDA in serum was dramatically lower than that in the control group (P 〈 0.01 ).②Swimming duration of rats in the experimental group was distinctly longer than that in the control group[(15.32±2.56) ,(8.50±2.15) rain, P 〈 0.05]. CONCLUSION:Bushen Yiqi Chinese herb can significantly improve the blood biochemical indexes of sport rats, enhance antioxidation, markedly increase the toleration of rats and delay the occurrence of sport fatigue.
