摘要
将β-环糊精、甲基-β-环糊精、羟丙基-β-环糊精、L-脯氨酸作为手性流动相添加剂,系统地研究了D,L-萘普生在RP-HPLC系统中的拆分。分别考察了手性流动相的种类,手性试剂羟丙基-β-环糊精的浓度,流动相的pH值,修饰剂的种类及浓度,三乙胺浓度和柱温等对拆分效果的影响,以HP--βCD为手性流动相添加剂,建立了HP--βCD手性流动相分离萘普生对映体的方法。结果表明:当流动相为25 mol/L HP--βCD、体积分数15%乙醇、体积分数0.5%三乙胺、pH3.5、柱温t25℃、流速V=1 mL/min时萘普生对映体得到了良好的基线分离,分离因子α可达1.29。
The enantiomeric resolution of naproxen was studied by reversed phase high performance liquid chromatography with hydroxypropyl-β-cyclodextrin (HP-β-CD) as a mobile phase additive. The effect of HP-β-CD concentration, organic modifier (type and concentration), pH of aqueous phase, percentage of triethylamine (TEA), column temperature, and different mobile phase additive on the resolution have been studied in detail, respectively. The optimal composition of mobile phase was 85:15 ( V/V) aqueous 0.5 % TEA, pH 3.5 (adjusted with glacial acetic acid)/ethanol containing 25 mmol/L HP-β-CD with the column temperature of 25 ℃ and the flow rate of 1.0 mL/min, it was found from the study on column temperature that the inclusion process was exothermic.
