摘要
实验采用HPLC法,以C18柱为色谱柱,CH3OH:H2O:HAc=80:16:4(含0.02 mol/L庚烷磺酸钠)为流动相,检测波长330 nm;对依替米星在生理盐水,5%葡萄糖,10%葡萄糖,复方氯化钠,葡萄糖氯化钠等溶媒中进行了稳定性研究,在室温(17℃),25℃,35℃情况下,将依替米星与不同的溶媒配伍后,在1 h,2 h,4 h,6 h,8 h分别测定它们的含量及pH值.结果表明依替米星在上述溶媒中的稳定性较好.在pH=2,pH=10,pH=12的条件下对依替米星进行稳定性研究,结果表明,依替米星在pH为2~10时稳定,当pH>10时依替米星被破坏.
The HPLC method has been used to investigate the stability of etimicin in 0.9 % sodium chloride, 5% glucose, 10% glucose, compound sodium chloride, glucose sodium chloride and so on. In the experiment, the ODS-C18 column (4.6 mm×250 mm), mobile phase: methanol/water/acetic acid=80/16/4 (containing heptane sulfonic sodium 0. 02 mol/L ), UV detection with wavelength of 330 nm were employed. After integrating each of the menstruums with etimicin, the content and pH value were measured after 1 h, 2 h, 4 h, 6 h and 8 h at a temperature of 17, 25 and 35℃, respectively. The result demonstrated that the stability of etimicin in these menstruums was good. Destructive tests have been made for etimicin under the conditions of pH=2, pH=10 and pH=12, respectively, and the result showed that etimicin was most stable at pH is 2-10 but destructed at pH〉10.
