结肠癌细胞上调脐静脉内皮细胞EMMPRIN的表达 被引量:2

摘要 目的:探讨结肠癌细胞对脐静脉内皮细胞中细胞外基质金属蛋白酶诱导因子(EMMPR IN)表达的影响,以及EMM-PR IN与肿瘤新生血管生成的关系。方法:建立结肠癌LoVo细胞与人脐静脉内皮细胞(HUVEC)共培养模型,并设HUVEC单独培养对照组,培养36 h后进行形态学观察,并应用免疫细胞化学方法检测内皮细胞EMMPR IN的表达。结果:共培养组HU-VEC形成大量管样结构,且EMMPR IN的表达明显强于HUVEC单独培养对照组强。结论:结肠癌细胞上调HUVEC EMMPR IN的表达,并诱导HUVEC管样结构的的形成,EMMPR IN的表达可能与肿瘤新生血管生成具有一定的相关性。 Objective To explore the effects of the colon carcinoma cell line LoVo on the expression of extracellular matrix metalloprotease inducer (EMMPRIN) in human umbilical vein endothelial cells( HUVEC), and to investigate the relationship between EMMPRIN and tumor angiogenesis. Method HUVEC were co-cultured with LoVo cells in transwell system. After 36 hours, phase contrast microscope and immunocytochemistry were used to detected morphological changes and the expression of EMMPRIN protein in HUVEC. Result By co-cultured with LoVo cells, HUVEC formed enormous tubular structures, at the same time, these HUVEC produced a lot of EMMPRIN protein comparing with the controls. Conclusion Colon carcinoma cell line LoVo can up-regulate the expression of EMMPRIN, and induce tubular structures in HUVEC. EMMPRIN may involved in the tumor angiogenesis.
