通鼻胶囊抗炎免疫药理作用的实验研究 被引量:2

摘要 目的观察通鼻胶囊对急、慢性炎症及免疫性炎症的药理作用,为临床应用和新药开发提供了科学的理论依据。方法通过建立多种实验性炎症的动物模型,观察药物疗效。同时对该药物抗炎免疫作用机理进行初步探讨。结果通鼻胶囊剂量依赖性地抑制DNCB所致小鼠迟发型超敏反应;对小鼠单核巨噬细胞系统吞噬功能有明显促进作用;剂量依赖性地抑制小鼠棉球肉芽组织增生,与阳性对照组相比无显著性差异;使小鼠胸腺、肾上腺减重,而对脾脏重量无明显影响;明显抑制组胺及5-HT引起的大鼠毛细血管通透性的增加;小鼠对通鼻胶囊的最大耐受量为人常用量的162倍以上。结论通鼻胶囊有对抗急、慢性炎症作用;有对抗免疫性炎症并可能具有免疫调节作用。通鼻胶囊的抗炎机制可能与下列因素有关:抑制炎症介质组胺、5-羟色胺的合成及释放,从而抑制炎症时毛细血管通透性的增高;抑制前列腺素E2、激肽的形成或其致炎活性;增强单核巨噬细胞系统(Mononuc lear phagocyte system,MPS)的吞噬功能。 Objective To study effects of Tongbi Capsule on inflammation, the purpose is to provide a scientific basis for clinical application and further research. Methods We observed the therapeutic effects of Tb on acute, chronic and immune inflammation in rats and mice by using several study models, and its mechanism is discussed preliminarily. Results The results showed that Tb significantly inhibited, in a dose- dependent manner, DTH induced by DNCB in mice. Tb significantly increased the phagocytic function of the mononuclear phagocyte system in mice. Tb dramatically inhibited the swelling of hind paws induced by carrageenin and the effective time lasted for 6 hours. Tb inhibited, in a dose -dependent manner, the proliferation of granuloma, the effect accompanied by thymic atrophy and adrenal atrophy, while the weights of spleen of rats were not affected. Tb markedly antagonized the increase of capillary permeability due to histamine and 5 - HT. Maximum tolerance dose of Tb is more than 162 times that of humans. Conclusion Tongbi Capsule has inhibitory action on acute, chronic and immune inflammation in rats and mice and its toxicity is very little. The mechanisms of its may be involved: inhibiting histamin, 5-HT, PGE2, and kinin by synthesis or release or activity, inhibiting cell-mediated immunity, and probably has immunomodulatory action.
