摘要
用细胞电穿孔融合仪,摸索最佳电融合参数,对5种分枝杆菌超声粉碎菌体混合抗原免疫的1只BALB/C小鼠脾细胞和NSl骨髓瘤细胞进行了电融合。在最佳电融合参数下,获得1.75个杂交瘤细胞克隆/105个脾细胞的最佳融合率。采用间接ELISA法筛选,经克隆化,获22株分泌抗分枝杆菌单克隆抗体(McAb)的阳性杂交瘤细胞株。并对其中11株进行了初步研究。
By using the electrofusion technique,the fusion of NSl myelona cells with spleen cells of BALB/c mouse immunized with the five species of mycobacteria was carried out.With the optimal electrofusion conditions,we obtained a high fusion rate of l.75 hybrid cell clones per l05 spleen cells.ELISA was used for screening secreting anti-mycobacteria McAb hybrid cells.After cloning,22 stable hybrid cells lines were selected,and 11 of them had been studied.
