摘要
本文介绍一种可产生单克隆抗体的杂交瘤细胞的电场诱导细胞融合技术。首先,施加非均匀交流电场(强度为250V/cm,频率为1MHz)或凝集素(植物血球凝集素、伴刀豆蛋白、链霉蛋白酶)使待融合细胞近接触;然后,施加三个电脉冲(强度为5kV/cm,脉宽为20μs)诱导细胞融合。经20天HAT选择培养,获得了52个大于10~4个以上杂交瘤细胞的大集落细胞团。
This paper introduces noyel technique of electrically induced cellfusion
to obtain hybridoma cells for production of monoclonal antibodies(McAbs).
After close contact betweell the cells was established by applying a non-
uniform alternating electric field (field strength: 250V/cm; frequency:
1MHz ) or some agglutinins (PHA, ConA or Pronase ), fusion of cells
was induced by applying three electric field pulses (field strength: 5KV/
cm, duration: 20μs ) to the cells. After 20 days of HAT selective incuba-
tion, 50 lumps of cells were obtained, each containing over 104 livillg hy-
bridoma cells.
