一种新的蛇毒解离素同系物的分离

Isolation of a Novel Disintegrin-like Homology from Agkistrodon halys Pallas Venom

摘要 为发现新的具有生物活性的蛇毒蛋白,以我国华南地区富有的蝮蛇蛇毒为分离样品源,采用高效液相色谱,从中分离出一种新的解离索同系物——AHP-4,建立了快速分离纯化蛇毒蛋白的方法。结构确认的结果表明,AHP-4的相对分子质量为7554 Da,是由一条多肽链组成的蛋白质,N-端第1~15位的氨基酸序列为GEECDCGSPANPCCD。经GeneBank protein-protein BLAST检索及人工检索,在已知蛋白质N-端第1~15位氨基酸序列中,未发现与AHP-4氨基酸结构完全相同的蛋白质。AHP-4N端第1~5位氨基酸序列与5种解离素中的氨基酸序列具有高度同源性,表明AHP-4是一种新的解离素同系物。 To took the venoms of Agkistrodon halys Pallas, which is abundant in South China, as the source of samples, a one step method for rapid isolation and purification of the snake venom extracts was established, for the purpose of the discovery of new polypeptide with anti-tumor activity. AHP-4 (the abbreviation of Akgistrodoon halys Pallas) which is a novel disintegrin-like homology from Agkistrodon halys Pallas venom was isolated by high performance liquid chromatography (HPLC). The molecular weight of AHP-4 was determined with electrospray ionization mass spectrum (ESI-MS). The molecular weight of AHP-4 was 7 554 Da. Under either reducing or non-reducing condition, AHP-4 appeared as a single band when analyzed by electrophoresis method. This result indicated that AHP-4 was a protein composed of a single peptide chain. The 1 st to the 15th amino acids at the N-terminal of AHP-3 were sequenced with the Edman degradation method. It is shown that the 1st to the 15th amino acids at the N-terminal end were GEECDCGSPANPCCD in sequence. After comparison of the amino acid sequence with that of the known protein sequence of protein-protein BLAST in GeneBank, it had not found any amino acid sequence which is identical with the 1st through 15th amino acids of AHP-4. Moreover, the 1~15 amino sequence of AHP-4 at the N-terminal shared high homology with 5 disintegrins, implying that AHP-4 was a novel disintegrin-like homology of disintngrins.
