摘要
对一株曲霉果糖转移酶菌株的产酶培养条件进行了研究。确定了最佳培养基组成:初始蔗糖质量浓度15~18g/dL,氮源为酵母膏,K2HPO4对果糖转移酶的产生具有明显的促进作用,添加0.2g/dLCMC能够使果糖转移酶活力提高到原来的1.3倍,在pH5.5,30℃条件下,果糖转移酶最高酶活力为30.42U/mL。HPLC分析结果表明,转糖基产物为总质量的55.8%。
The optimal conditions for fructosyltransferase production from Aspergillus sp. were investigated. The initial concentration of sucrose was 15~18 g/dL. Yeast extract was the best nitrogen source. K2 HPO4 was effective in increasing enzyme production. The enzyme activity was increased 30% by addition of 0. 2 g/dL of CMC in the medium. The highest fructosyltrnsferase activity was 30.42 U/mL at pH 5. 5 and 30 ℃. Fructooligosaccharides was 55.8% of the total products.
