The Association between Graves' Disease and a C/T Single-Nucleotide Polymorphism in the CD40 Gene

摘要 目的:探讨CD40基因5′非翻译区域C/T多态性和Graves病(GD)甲亢发病的相关性。方法:采用序列特异性聚合酶链式反应-限制性内切酶酶切长度多态性(PCR-RFLP)方法分析106例GD甲亢患者和106例正常对照者CD40基因5′非翻译区域C/T多态性。结果:GD甲亢患者CC基因型明显高于正常对照组(49.1%vs32.1%),C等位基因频率明显高于正常对照组(63.7%vs50.5%)。结论:CD40基因5′非翻译区域的C等位基因和GD甲亢相关。 Objective: To investigate the C/T polymorphism in the 5′-untranslated region(UTR) of the CD40 gene and its relationship with Graves'disease. Methods: The C/T polymorphism in the CD40 gene 5′ UTR in one hundred and six GD patients and one hundred and six normal patients (normal) was analyzed by using polymerase chain reaction-restrictionfragment length polymorphism (PCR-RFLP). Results: The CC genotype of GD patients was significantly higher than that in the normal subjects (49.1% vs 32.1%),on the other hand, the frequency of the C allele was significantly higher than that in the normal subjects (63.7% vs 50.5%). Conclusion: The C allele in the 5′-untranslated region of the CD40 gene may be associated with etiological factor of GD.
