缺氧对肺动脉内皮细胞生长特性、细胞内游离钙及cAMP浓度的影响 被引量:4

The effect of hypoxia on cAMP and calcium concentrations in cultured pulmonary artery endothelial cells

摘要 作者观察了缺氧状态下培养的小牛肺动脉内皮细胞(PAEC)的形态、生长曲线和蛋白质含量的变化,动态测定了缺氧时PAEC细胞内游离钙浓度([Ca(2+]i)和cAMP含量。结果发现:(1)PAEC能在2%O_2,缺氧条件下生长,其光镜形态、生长曲线和细胞蛋白质含量与21%O_-2常氧条件下的相似;(2)向PAEC的无钙细胞悬液内分别通入氮气、3%O_2和5%O_21~20min,细胞内([Ca(2+]i)均明显升高;(3)通入氮气1min,细胞内cAMP含量明显上升,继续缺氧则cAMP有向基础值接近的趋势。上述结果提示:培养的PAEC对缺氧有较强的耐受力,缺氧可直接影响PAEC的第二信使传递系统。 The effects of hypoxia on cell growth, cAMP and cytosolic free calcium concentrations([Ca(2+)]i)of cultured bovine pulmonary arterial endothelial eells(PAEC)wereevaluated.The results obtained were as follows:(1)There were not much differenees in cellgro wth characteristic and eell pro tein co ntent between hypo xic(2%O2)and normoxic(21%O2)groups.It seems that hypoxia didn’t cause PAEC hypertrophy or hyperplasia,(2)[Ca(2+)]i of PAEC were determined by fluorescent probe Fura-2,and cAMP by RIAassay.Changes of [Ca(2+)]i were noticed in PAEC suspended in calcium-free medium exposedto 0% O2(pure N2),3%O2 and 5 %O2 hypoxic challenges respectively for 1 to 20 minutes。[Ca(2+)]i in PAEC under hypoxia elevated significantly and continuously up to 400~500nmol/L(P<0.00 1).The coneentration of cAMP increased significantly(P < 0.01)and transiently atthe cnd of 1 min hypoxic challenge (0% O2).These results suggested that hypoxia could ex-ert direct influences upon second messenger system in cultured PAEC.
