摘要
目的探讨大肠杆菌中表达嗜肺军团菌的热休克蛋白60(HSP60)的表达。方法利用聚合酶链式反应(PCR)技术扩增嗜肺军团菌HSP60基因,将其定向插入载体pUC18构建重组原核表达质粒,重组质粒转化到大肠杆菌JM109后IPTG诱导表达,并对其表达产物进行SDS-PAGE和Western-blotting分析。结果重组质粒在大肠杆菌中成功表达,可被抗军团菌抗血清识别。结论HSP60基因可在大肠杆菌中表达,并有免疫原性。
[Objective] To study the heat shock protein 60 gene of Legionella pneumophila in E.coli for further research of DNA vaccine of legionella. [Methods] The HSP60 gone of Legionella pneumophila was amplified by PCR, then inserted into pUC18 vector to construct recombinant plasmid. And then, the recombinant plasmid was transferred into E.coli JM109 and induced with IPTG. The expression product of the gene was analyzed by SDSPAGE and Western-blotting. [Results] The recombinant plasmid could express in E.coli. And the recombinant HSP60 protein was recognized by antiserum against legionella pneumophila. [Conclusion] HSP60 gene can be expressed in E.coli and the recombinant protein has immunocompetence.
