梅毒患者外周血树突状细胞亚型及血清IL-12I、FN-γ的检测 被引量:1

摘要 目的探讨树突状细胞(DC)各亚型及白细胞介素12(IL-12)、干扰素-γ(IFN-γ)在梅毒发病机制中的作用。方法流式细胞仪测定23例梅毒患者和15例正常人的外周血DC各亚型的表达;酶联免疫吸附试验(ELISA)方法检测其血清IL-12和IFN-γ的水平。结果23例梅毒患者外周血髓系树突状细胞亚型(CD11c+)的表达为(1.51±2.04)%,与15例正常对照组(0.38±0.18)%比较显著升高(P<0.01),梅毒患者外周血淋巴系树突状细胞亚型(CD123+)的表达为(0.37±0.21)%,与正常对照组(0.38±0.22)%比较无显著差异(P>0.05)。梅毒患者血清中IL-12的水平为(52.70±103.04)pg/mL,与正常对照组(0.98±1.47)pg/mL比较显著升高(P<0.05);梅毒患者血清中IFN-γ的水平为(6.12±9.76)pg/mL,与正常对照组(0.25±0.18)pg/mL比较显著升高(P<0.01)。梅毒患者外周血CD11c+、CD123+百分数与血清中IL-12、IFN-γ的水平不存在相关关系(P均>0.05)。结论梅毒螺旋体(TP)进入体内后主要诱导CD11c+型DC表达,引发Th1反应,启动细胞免疫;IL-12及IFN-γ参与了TP进入体内后的免疫过程,可能在TP的清除过程中发挥重要的免疫调节作用;DC各亚型的表达与IL-12、IFN-γ的水平不存在相关关系。 Purpose To investigate the role of dendritic cells (DC) and interleukin-12(IL-12),interferon-gamma(IFN-γ) in the pathogenesis of syphilis. Methods Flow cytometry was used to detect DC subsets in peripheral blood and ELISA method was used to detect the serum level of IL-12, IFN-γ in 23 patients with syphilis and 15 healthy controls. Results The expression of CD11c^+ DC was (1.51 ± 2.04) % in peripheral blood in syphilis patients, increased more significantly than that in normal controls (0.38 ± 0.18) % (P〈0.01). The expression of CD123^+ DC in peripheral blood in syphilis patients was (0.37 ± 0.21) %, with no significant difference compared with that in controls(0.38 ± 0.22)% (P〉0.05). The serum level of IL-12 was (52.70 ± 103.04)pg/mL, increased more significantly than that in normal controls (0.98 ± 1.47)pg/mL (P〈0.05) and level of IFN-γ in syphilis patients was(6.12 ± 9. 76)pg/mL, increased more significantly than that in normal controls(0.25 ± 0.18)pg/mL(P〈0. 01). There was no correlation between the expression of DC subsets and the level of IL-12 or IFN-7 (P〉0.05). Conclusions CD11c^+ DC and initiated cell-mediated immune Treponema pallidum induced mainly the expression of response. IL-12 and IFN-γ might take part in the immune processes after Treponema pallidum entering into body, and play a prominent role in Treponema pallidum clearance. The expression of DC subsets had no correlation with the level of IL-12 and IFN-γ.
