摘要 目的:探讨雷公藤内酯醇(TP)对小鼠巨噬细胞表达血管内皮生长因子(VEGF)的抑制作用。方法:以脂多糖(LPS)激活的小鼠腹腔巨噬细胞为研究对象,利用RT-PCR、免疫组化检测不同剂量雷公藤对激活巨噬细胞VEGFmRNA表达及VEGF合成的影响。结果:TP可以抑制LPS诱导的巨噬细胞VEGFmRNA表达及VEGF合成,并呈剂量依赖性。结论:TP抑制巨噬细胞VEGFmRNA转录及VEGF合成可能是其治疗子宫内膜异位症的作用机理之一。 Objective: To investigate the effect oftriptolide on vascular endothelial growth factor (VEGF) expression in murine macrophages, and explore the therapy mechanism of triptolide on endometriosis. Methods: The peritoneal macrophages of mice were used in this study. The effects of triptolide on VEGF mRNA expression, and intracellular protein production and secretion induced by LPS were measured by RT-PCR and immunohistochemical method. Results: LPS of 10 μg/ml significantly increased VEGF mRNA expression and intracellular production of VEGF in macrophages, while triptolide inhibited the effects of LPS in a dose-dependent manner. Conclusion: Triptolide inhibits VEGF expression and production of macrophages, suggesting that the inhibitory influence of triptolide on VEGF expression and production may be one of the mechanisms underlying its therapy effect on endometriosis.
