摘要
目的:探索血小板糖蛋白(glycoprotein,Gp)ⅡbⅢa复合体的基因多态性是否与动脉硬化性脑血栓形成有关。方法:GpⅡb组病例122例,对照组99例,GpⅢa组病例127例,对照组104例,提取空腹静脉血白细胞中DNA,经聚合酶链反应(PCR)后,分别用限制性内切酶FokⅠ、ScrFⅠ酶切,2.2%琼脂糖凝胶电泳,紫外分析仪观察。数据统计应用SPSS11.5forWindows统计软件包。结果:GpⅡb组中≤70岁的人GpⅡbSer843等位基因的纯合子型与增加动脉硬化性脑血栓形成的危险相关(P=0.029);将病例组和对照组中的男性,按照基因型bb组和(aa+ab)组进行分析,结果显示两组间有显著差异(P=0.01,OR=2.194,95%CI=1.177～4.091);GpⅢa组共231例均为GpⅢaPlA1基因型。结论:在动脉硬化性脑血栓形成中有GpⅡbIle/Ser843的基因多态性异常;而GpⅢaPlA1基因型无异常。
Aim： To explore the relationship between the GpⅡb-Ⅲa polymorphism and arteriosclerotic cerebral thrombosis in Chinese. Methods： Gp lib genotype was investigated in 122 patients with arteriosclerotie cerebral thrombosis and 99 eontrol subjects. GpⅢa genotype was studied in 127 patients with arteriosclerotic eerebral thrombosis and 104 control subjects. Peripheral venous blood samples were drawn from all subjeets after overnight fasting to extract genomic DNA, the GpⅡb genotype was determined from leukocyte DNA by polymerase chain reaetinn （PCR） followed by Fok Ⅰ digestion; and the Gp Ⅲa genotype was determined from leukocyte DNA by polymerase ,hain reaction （PCR） followed by Serf Ⅰ digestion. After 2.2% agarose gel electrophoresis with ethidium bromide staining, the products were analyzed with ultraviolet anal）zer. All statistieal analyses were performed with SPSS 11.5. Results： In the Gp Ⅱb system aged ≤ 70 years, homozygosity for the Ser^843 allele of the glycoprotein Ⅱb was associated with increased risk of arteriosclerotic cerebral thrombosis （ P = 0.029）, and in the study of men （ P = 0.01, OR = 2. 194, 95% CI = 1. 177～4.091 ） [ bb vs aa ＋ ab].Logistic regression analysis confirmed the result above. All donors in the GpⅢa （n = 231） expressed the GpⅢa Pl^A1 phenotype. Conclusion： The GpⅡb Ile/Ser^843 gene polymorphism is abnormal in the arteriosclerotic cerebral thrombosis, and the GpⅢa Pl^A1 phenotype is normal in the arteriosclerotic cerebral thrombosis.
