血小板糖蛋白Ⅱb的基因多态性与脑梗死的相关性研究 被引量:6

摘要 目的从分子病因学的角度探索血小板糖蛋白(Glycoprotein,GpⅡb)的基因多态性是否与脑梗死的发生相关,为缺血性卒中的预防及进一步治疗提供理论基础.方法选取病例组122例,对照组99例,提取空腹静脉血白细胞中DNA,经聚合酶链反应(PCR)后,用限制性内切酶Fok I酶切,2.2%琼脂糖凝胶电泳,紫外分析仪观察.结果 GpⅡb组中≤70岁的人GpⅡbSer843等位基因的纯合子型与增加脑梗死的危险相关(P=0.029);将病例组和对照组中的男性,按照基因型bb组和(aa+ab)组进行分析,结果显示两组之间有显著差异(P=0.01,OR=2.194,95%CI=1.177~4.091).结论 GpⅡb Ⅱe/Ser843的基因多态性与脑梗死的发生相关. Objective To explore the relationship between polymorphism of platelet glycoprotein(Gp) Ⅱ b and cerebral infarction in Chinese at the aspect of molecular etiology, and to provide the theoretical basis for prevention and therapy of ischemic stroke. Methods Gp Ⅱ b genotype was investigated in 122 patients with cerebral infarction and 99 control subjects. Peripheral venous blood samples were drawn from all subjects after overnight fasting to extract genomic DNA, the Gp Ⅱb genotype was determined from leukocyte DNA by polymerase chain reaction(PCR) followed by Fok I digestion. After 2.2% agarose gel electrophoresis with ethidium bromide staining, the products were analyzed with ultraviolet analyzer. All statistical analyses were performed with SPSS11.5. Results In the Gp Ⅱb system aged ≤70 years, homozygosity for the Ser 843 allele of Gp Ⅱb was associated with increased risk of cerebral infarction (P = 0. 029 ) , and in the study of men ( P = 0.01, OR = 2. 194, 95% CI = 1. 177 ~ 4.091 ) [ genotype bb vs aa + ab]. Conclusions The Gp Ⅱ b Ⅱe/Ser843 gene polymorphism is associated with cerebral infarction.
