摘要
目的:研究植酸对人胃癌细胞的生长抑制作用及机制。方法:采用MTT实验、Hoechst33342荧光染色法观察不同剂量植酸对SGC-7901细胞增殖的影响,免疫组化法检测Bcl-2蛋白的表达。结果:植酸对SGC-7901细胞增殖具有抑制作用,且存在剂量-效应关系。荧光染色可见,植酸作用组细胞出现典型的细胞核浓缩、边集、裂解的凋亡特征。植酸处理组细胞中Bcl-2蛋白表达较对照组降低。结论:植酸对SGC-7901细胞增殖具有抑制作用和诱导凋亡作用。
Objective: To examine the action of phytic acid on proliferation inhibition of SGC-7901 cells. Methods: We observed the effect of phytic acid on proliferation of SGC-7901 cells by MTT experiment and Hoechst33342 fluorescence staining, and the expression of Bcl-2 was determined by immune histochemisty method. Results: The proliferation of SGC-7901 cells was inhibited by phytic acid with dose-dependent manner. From fluorescence staining, we detected nuclei concentration, aggregation to edge and splitting in the phytic acid group. The expression rate of Bcl-2 protein in phytic acid group was lower than that of control group. Conclusion: Phytic acid has the effect of proliferation inhibition on SGC-7901 cells, and can induce the cells to apoptosis. [ACTA NUTRIMENTA SINICA, 2006, 28(1): 51-53 ]
