摘要 目的:研究胸腺肽α1(Tsα1)在诱导肿瘤细胞凋亡中的作用。方法:应用MTT比色分析法研究Tsα1对人肝癌SMMC7721细胞株的细胞毒作用。流式细胞仪检测法分析Tsα1作用后处于G1期SMMC7721细胞的比率和对凋亡抑制基因bcl2蛋白的抑制率。结果:Tsα1(4000μg/mL)体外培养24、48和72h,对SMMC7721肿瘤细胞抑制率分别是(1.92±0.02)%、(3.27±0.03)%和(7.69±0.03)%,P<0.005;凋亡率分别是(2.5±0.1)%、(18.3±0.2)%和(11.6±0.1)%,P<0.001;bcl2蛋白的抑制率为(1.35±0.1)%、(2.2±0.01)%和(5.53±0.01)%,P<0.005,并呈现细胞凋亡的形态学改变,具有剂量和时间依赖性。结论:Tsα1可直接抑制人肝癌SMMC7721细胞株增殖,抑制bcl2基因表达和诱导肿瘤细胞凋亡。 OBJECTIVE: To stude the apoptosis induced by thymope α1 (Tsαl) in human hepatoeelluar cancer SMMC-7721 cells. METHODS: Tsα1 inhibitatory SMMO7721 cell's growth by was studied MTT assay; morphological characteristics of apoptosis was observed by microscopy; cell apoptosis rate was measured by flow cytometry and agarrose gel electronphoresis; expression of bcl-2 was detected by flow cytometry. RESULTS: When the concentration of Tsα1 was 4 000 μg/mL, the inhibitatory rates of SMMC-7721 cells were (1.92±0. 02)%, (3.27± 0.03)% and (7.69±0.03)% at 24 h, 48 h and 72 h, respectively,P〈0.005. Theapoptosis rates were (2.5±0.1)%, (18. 3± 0.2)% and (11.6±0. 1)% at 24 h, 48 h and 72 h, respectively, P〈0. 001. The inhibitatory rates of bel-2 expression were (1.35 ±0. 1)%(2.2+0.01)% and (5.53+0.01)% at 24 h, 48 h and 72 h, respectively,P〈0. 005. The morphological changes of ap optosis were fuound. CONCLUSION, Tsα can inhibit prolifera tion of SMMC-7721 cells, induce apoptosis in SMMC 7721 cells and inhibit expression of bcl 2.
