摘要
通过500多头次采用母鸭诱情法采精和食指定位法输精,对番鸭本品种人工授精方法、输精量、输精间隔时间、输精时间以及稀释液对受精率影响等进行系列研究.结果表明:番鸭平均采精量为1.07±0.26mL只-1,密度为(1.85±0.23)×109mL-1,活力为7.5-9.8;以16:00-17:00输精,输精量0.05mL只-1,间隔6d输精1次为好,受精率可达89.11%;采用人工授精,公鸭利用率比自然交配提高20倍,可大幅度提高饲养效益;用不同稀释液(生理盐水、蛋黄葡萄糖液和磷酸缓冲液)按11稀释番鸭精液与原精液输精比较,以磷酸缓冲液稀释组的受精率(90.01%)较高,但与原精组受精率(89.09%)差异不显著(P>0.05);用磷酸缓冲液和Lake液以11稀释后在2-5℃保存24h输精,受精率分别为30.56%和45.32%
A series of artificial insemination techniques of Muscovy ducks including methods, doses, intervals, times and diluents to be used as well as their influences on the fertility were studied. Semen was collected by the method of female teaser and insemination was operated with the help of index finger to guide the injector to the vagina of female duck, more than 500 ducks in different times were used for test. The results showed that the average volume of semen collected from each drake at each time was 1.07±0.26 mL; the sperm concentration (1.85±0.23)×10 9·mL -1 , and the sperm motility score 7.5-9.8, making artificial insemination at 16:00-17:00 o′clock with the dose of 0.05 mL pooled semen and repeating AI after 6 day interval seem to be the best choice and the fertility can reach 89.11%. The utilization rate of a drake for AI was 20 times as high as that of a drake for natural mating, hence, the feeding efficiency was greatly increased. At the same time, three diluents normal saline solution, yolk glucose solution and phosphate buffer solution (PBS) used as semen extenders in a ratio of 1∶1 were compared. The highest fertility reaching 90.01% was found in the group of using dilute PBS, however as it was compared with the control, in which the semen used for AI was undiluted and the fertility was 89.09%, the difference was not significant ( P >0.05); when the semen was diluted by PBS or Lake′s solution in a ratio of 1∶1 and stored at 2-5 ℃ for 24 hours before AI, the fertility obtained was 30.56% and 45.32% respectively.
