谷物样品中各单糖及游离糖、淀粉、可溶性和不溶性NSP流水序列测定方法:用80%乙醇提取游离糖;而后用乙酸缓冲液、α-淀粉酶和葡萄糖苷酶分解淀粉,得到可溶和不可溶NSP;用葡萄糖氧化酶/过氧化酶方法测定淀粉;NSP和游离糖再经水解。与内标共同完成还原和酰化过程,用装有高级性毛细管柱的GC测定。该法不仅可给出构成NSP各单糖含量,也可给出淀粉和游离糖的量值。
A stream-lined assay procedure has been developed for the determination of monosaccharides, free sugars,starch, soluble and insoluble polysaccharides in cereal and cereal products on a single-sample basis. The sample is extracted with 80% ethanol to remove free sugars, and then treated with highly specific α-amylase and amyloglucosidase (in acetate buffer at pH 5. 0)to degrade starch and to fractionate the soluble and insoluble non-starch polysaccharides (NSP). An aliquot of the glucose-containing supernatant is taken and analyzed for starch ,using the glucose oxidase/peroxidase method. The free sugars, the soluble and insoluble fractions of NSP are hydrolyzed by acids, and the released monosaccharides are reduced and acetylated along with a known amount of internal standard. The resulting alditol acetates are determined by GC with a polar capillary column. The proposed method can determine not only the monosaccharide constituents of NSP ,but the starch and free sugars as well.
