用基底动脉结扎法造成犬脑干缺血.在缺血前3h十二指肠灌注给予绞股蓝皂甙(150mg·kg^(-1)).脑干缺血后6h的观察表明,绞股蓝皂甙减轻神经元缺血性损害(光镜及电镜),改善脑干听觉诱发电位(BAEP)的异常变化,降低缺血导致的磷脂酶A_2(PLA_2)活性升高,增强超氧化物岐化酶(SOD)活性.结果表明绞股蓝皂甙对脑干缺血性损伤有保护作用,其机制可能与其拮抗自由基和钙相关性损伤有关.
The brainstem ischemic injuries were produced by the occlusion of the basilar artery for 6 h in dogs. Gypenoside (150 mg ?kg-1)were intradudinally given at 3 h before the occlusion. We discovered that gypenoside alleviated the ischemic neuronal damage (histopathologically evaluated by the light and electronic microscope), ameliorated the abnormal changes of brainstem auditory evoked potentials (BAEP) (F<0. 01), inhabited the phos-pholipase A2( PLA2)activation induced by the ischemia (P<0. 01) and intensified the superoxide dismutase (SOD) activity (P<0.01). The results suggest that gypenoside has the protective effects on brainstem ischemic injuries and its mechanism may involve its capacity to antagonize the Ca2+ and free radicals related injuries.
