一氧化碳下调高血压大鼠血管平滑肌细胞硫化氢体系 被引量:4

Study on Carbon Monoxide Down-Regulating Hydrogen Sulfide Pathway of Vascular Smooth Muscle Cells in Hypertensive Rats

摘要 目的研究一氧化碳(CO)对自发性高血压大鼠(SHR)血管平滑肌细胞(VSMC)中硫化氢(H2S)/胱硫醚γ裂解酶(CSE)的影响。方法SHRVSMC分为氯化血红素(hemin)实验组和hemin对照组,硫电极法测定培养上清中H2S含量,竞争性RTPCR法检测细胞中CSEmRNA水平。结果实验组较对照组培养上清中H2S含量显著降低[(16.03±2.15)μmol/Lvs(13.31±1.74)μmol/L],CSEmRNA水平较对照组降低[(0.17±0.04)vs(0.13±0.03)]。结论CO可下调SHR的VSMC中H2S/CSE体系。 Objective To investigate the effect of carbon monoxide(CO) on hydrogen sulfide(H2S)/cystathionine-γ-lyase system(CSE) in vascular smooth muscle cells(VSMC) of spontaneous hypertensive rats(SHR). Methods The SHR VSMC were divided into 2 groups:experimental group( hemin was added to the culture media at a concentration of 10 μmol/L)and control group (dimethyl sulfoxide was added to the culture media at an equal volume). The content of H2S in the cultrue media was measured by sulfide electrode method, and the CSEmRNA level was assayed by competitive reversed transcription - polymerase chain reaction(RT PCR). Results Compared with control group, the content of H2S in the culture media of hemin treated SMCs was significantly lower[ ( 16.03 ± 2.14) μmol/L vs ( 13.31 ± 1.74)tmaol/L], and the CSEmRNA level in hemin treated SMC,s was decreased markedly(0.17 ± 0.04 vs 0.13 ± 0.03 ). Conclusion CO can down regulate the H2S/CSE system in SMC of SHR.
