摘要 目的:探讨细胞外信号调解激酶(ERK)对蟾蜍灵诱导白血病细胞凋亡的影响。方法:形态学观察和流式细胞仪检测细胞凋亡。结果:0.5μmol/L蟾蜍灵作用K562细胞24 h诱导典型的细胞凋亡,预先用ERK抑制剂PD98059处理细胞1 h,明显增加蟾蜍灵诱导的细胞凋亡率(P<0.05);而用佛波酯预先处理后,明显拮抗蟾蜍灵的作用(P<0.05)。结论:ERK负向调节蟾蜍灵诱导的K562细胞凋亡过程。 Objective: To explore the effect of extraceUular signal-regulated kinase (ERK) on the apoptosis induced by bufalin in K562 ceils. Methods: Apoptosis was determined by morphological method and flow cytometry. Results: Bufalin of 0.5 μmol/L induced the apoptosis of K562 ceils at 24 hours, and the action of bufalin was enhanced by ERK inhibitor PD98059 and reduced by 12-O-tetradecanoylphorbol-13-acetate. Conclusion: ERK negatively regulated the apoptosis induced by bufalin in K562 cells.
