摘要 目的研究蟾蜍灵诱导结肠癌细胞的凋亡作用,并探讨其治疗结肠癌的作用机制。方法:以不同浓度的蟾蜍灵处理结肠癌SW620细胞后,采用四唑蓝比色试验检测癌细胞增殖,分别采用光镜和琼脂糖凝胶电泳检测癌细胞凋亡情况,采用Fura-2荧光复合技术测结肠癌细胞内[Ca2+]i浓度。结果:蟾蜍灵对结肠癌细胞增殖具有很强的抑制作用。蟾蜍灵能够诱导结肠癌细胞凋亡。在结肠癌细胞凋亡过程中,胞内[Ca2+]i明显升高,以凋亡早期更为明显。1h时为288.56±8.46nmol/L,明显高于对照组(125.56±5.25)(P<0.01)。结论:蟾蜍灵对结肠癌细胞生长具有很强的抑制作用,与诱导凋亡有关。Ca2+在蟾蜍灵诱导结肠癌细胞凋亡过程中起重要的作用。 Purpose To explore the mechanism of bufalin on colon carcinoma SW620 cell line. Methods, After colon cancer cells were treated with different dose of bufalin,we studied the growth using MTT,and apoptosis using light and agarese gel electrophoresis, and [Ca2+] change using Fura-2 fluorescin load method. Results Bufalin had a remarkable supression effect on colon carcinoma SW620 cell line. Anticancer effect of SW620 would be correlation with apoptosis. [Ca2+] i was significantly increased, especially at the initial stage of apoptosis, it is 288.56 ± 8,46 nmol/L at 1 h, which was significantly control(125,56 ± 5,25)(P〈0.01 ). Conclusion Bufalin can inhibit the proferation of human colon carcinoma cell line SW620.The induction of apoptosis would be a very Keyimportant mechanism in the treatment of colon carcinoma. [Ca2+]i play an important role in the process of bufalin induced spoptosis of colon carcinoma cells.
