摘要
目的研究姜黄素诱导人乳腺癌MCF-7细胞凋亡的条件及机理。方法用MTT法测定姜黄素抑制MCF-7细胞增殖的IC50值;流式细胞仪测定姜黄素处理后MCF-7细胞周期分布变化与凋亡量;光镜观察MCF-7细胞形态学变化;RT-PCR技术测定MCF-7细胞bcl-2、Bax mRNA表达量变化。结果姜黄素抑制MCF-7细胞增殖的IC50值为18.5μM;在IC50值条件下,MCF-7细胞凋亡率为8.9%,可见明显的核固缩与胞浆浓缩,bcl-2mRNA表达明显降低(P<0.01),Bax mRNA表达明显增高(P<0.01)。结论姜黄素可通过抑制bcl-2、促进Bax mRNA表达诱导MCF-7细胞凋亡,并有效抑制其增殖。
Objective To study the condition and mechanism of the apoptosis of human breast cancer cell MCF-7 induced by curcumin. Methods MTT method was used to assay the IC50 value of curcumin and its antiprohferation effect. FCM was accepted for detecting of cell cycle distribution and apoptosis. The morphological changes of apoptotic cells were evaluated by light mircroscopy. The mRNA expression levels of bcl-2 and Bax were analysed by using RT-PCR technique. Results The IC50 value of curcumin inhibiting the proliferation of MCF-7 cell was 18.5μM,and under that condition,the apoptotic rate of MCF-7 cells was 8.9% ,obvious cytoplasmic shrinkage and nuclear condensation could be seen, and the mRNA expression of bcl-2 and Bax significantly decressed and increased (P〈0. 01) respectively. Conclusion Curcumin may induce apoptosis of MCF-7 cell and inhibit its proliferation through inhibiting bcl-2 and promoting Bax mRNA expression.
